DNA immobilization on surfaces is one of the most promising tools to achieve site-specific functionalization of biomaterials. We studied the adsorption of charged DNA fragments on positively charged gold nanoparticles. The results obtained using density functional theory are in excellent agreement with experimental data. We performed a molecular dynamics simulation of a model system, made of 16 DNA double strands and 10 gold nanoparticles. At 25 degrees C, the dynamic behavior of the system suggests that the DNA fragments cover, in average, 4.7 gold nanoparticles. It has also been possible to determine the orientation of the individual DNA fragments on the surface, showing that the orientation of the DNA fragment is strongly influenced by the surface.High level expression of functional human IL-4 receptors on the lymph node lymphocytes.
The human T cell line, RJ2.2.8, constitutively expresses receptors for IL-4 on its surface. To investigate whether IL-4 can exert its effect through these receptors, we studied the binding of IL-4 to the lymph node lymphocytes from healthy donors by autoradiography. This was compared with the binding of IL-4 to the RJ2.2.8 line. The binding to the lymph node lymphocytes was specific and saturable. The binding of [125I]IL-4 to lymph node lymphocytes was inhibited by excess of unlabeled IL-4. The Scatchard analysis gave a curvilinear plot, indicating the presence of multiple classes of binding sites. The relative equilibrium binding constants were of the same order of magnitude as that of the RJ2.2.8 line. Thus, both the lymph node lymphocytes and the RJ2.2.8 cell line possess functional IL-4 receptors.
